Endocannabinoids are a class of lipid-based neurotransmitters that are synthesized on demand in the body. They play a crucial role in the endocannabinoid system, which is involved in regulating a wide range of physiological processes. The term "endocannabinoid" itself is derived from "endogenous," meaning originating within the body, and "cannabinoid," referring to compounds that interact with cannabinoid receptors.

The two most well-studied endocannabinoids are anandamide (AEA) and 2-arachidonoylglycerol (2-AG). Anandamide, often referred to as the "bliss molecule," is believed to be involved in feelings of well-being and pleasure. 2-arachidonoylglycerol is present in higher concentrations in the brain and is thought to be involved in regulating pain, appetite, and mood.

These molecules are not stored in vesicles like traditional neurotransmitters. Instead, they are produced by postsynaptic neurons in response to a signal and then diffuse across the synapse to bind with cannabinoid receptors (CB1 and CB2) located on the presynaptic neuron. This retrograde signaling mechanism allows endocannabinoids to modulate the release of other neurotransmitters.

The endocannabinoid system's functions are extensive and include regulating appetite, pain sensation, mood, memory, immune function, and stress response. Its influence on these diverse systems highlights its importance in maintaining homeostasis. Research into endocannabinoids and their therapeutic potential is ongoing, with implications for treating conditions such as anxiety, depression, chronic pain, and neurodegenerative diseases.
