Emaljorganet, or the enamel organ, is the epithelial component of the developing tooth germ that is responsible for enamel formation. It arises from the oral epithelium as part of the cap stage of tooth development, together with the dental papilla and the dental sac. The enamel organ is organized into four histological layers: the outer enamel epithelium, the inner enamel epithelium, the stellate reticulum, and the stratum intermedium. The inner enamel epithelium differentiates into ameloblasts, the enamel-secreting cells.

During tooth development, the enamel organ interacts with the surrounding mesenchyme of the dental papilla to coordinate odontogenesis. Dentin is formed first by odontoblasts derived from the dental papilla, and subsequently enamel is deposited by ameloblasts derived from the inner enamel epithelium. The enamel matrix produced by ameloblasts becomes mineralized to form mature enamel.

In the bell stage the enamel organ undergoes histodifferentiation and morphogenesis, shaping the future crown. After enamel formation is complete, the enamel organ largely degenerates, leaving the reduced enamel epithelium that covers the enamel and participates in the eruption process by creating the eruption pathway.

Clinical relevance is linked to disturbances in enamel organ development or function. Abnormal enamel organ activity can lead to enamel defects such as amelogenesis imperfecta, or other disturbances in enamel formation, often with genetic or environmental contributions affecting ameloblast differentiation and activity.
