Checkpointmediated is a term used to describe regulatory processes that are governed by cellular checkpoints. These checkpoints act as surveillance points within biological systems, monitoring for errors or perturbations and delaying progression until the problem is resolved. The concept encompasses both the maintenance of genome integrity during the cell cycle and the coordination of signaling networks in response to stress.

In eukaryotic cells, checkpointmediated regulation is most clearly illustrated in the cell cycle, including DNA damage checkpoints (G1/S, S, and G2/M) and the spindle assembly checkpoint. Sensor kinases such as ATM and ATR detect damage or replication stress and activate transducers like CHK1 and CHK2, which then modulate effectors such as p53, p21, Cdc25, and Wee1. These interactions control the activity of cyclin-dependent kinases and the anaphase-promoting complex/cyclosome (APC/C), enforcing cell cycle arrest, promoting DNA repair, or triggering senescence or apoptosis if damage is irreparable.

Beyond cell cycle control, checkpointmediated regulation can influence other signaling pathways that rely on checkpoint-like surveillance to maintain homeostasis, such as responses to replication stress, chromosomal instability, or developmental cues. Defects in checkpointmediated pathways are associated with genomic instability and cancer, highlighting their central role in cellular reliability.
