Biopsy methods include core needle biopsy, punch biopsy, incisional or excisional biopsy, and fine-needle aspiration. The choice of method depends on the target organ, lesion characteristics, and diagnostic goals. Specimens may be fixed in formalin and embedded in paraffin (FFPE) for histology and immunohistochemistry, or preserved fresh-frozen for nucleic acid analysis and molecular testing. Processing steps and quality considerations, such as cellularity, tumor content, and avoidance of contamination, strongly influence downstream analyses.

Applications span diagnostic and research contexts. In clinical settings, biopsy-derived material supports pathology diagnoses, infectious disease identification, and molecular profiling to guide therapy, including cancer genomics and biomarker testing. In research, biopsy-derived cells can be used to establish organoids or cell lines, study tumor heterogeneity, or investigate disease mechanisms.

Limitations include the invasive nature of obtaining material, sampling bias, and potential artifacts from fixation or processing. Ethical considerations cover informed consent, patient privacy, and governance of biobanked biopsy material.
