VirB1VirB11

virB1virB11 refers to the gene cluster encoding the VirB1 through VirB11 components of a Type IV secretion system (T4SS) found on some bacterial plasmids, most famously the Ti (tumor-inducing) plasmids of Agrobacterium tumefaciens. This secretion system mediates transfer of DNA and protein effectors across the bacterial membranes and into host cells, plays a central role in plant infection, and serves as a widely studied model of T4SS architecture.

The VirB proteins assemble into a trans-envelope apparatus that spans the inner and outer membranes and, in some contexts, extends pilus-like structures. VirB1 encodes a lytic transglycosylase that remodels peptidoglycan to permit assembly of the complex. VirB2 encodes the major pilin forming the T-pilus. VirB3 is a small inner membrane protein, while VirB4 and VirB11 are ATPases providing the energy for secretion; VirB11 acts at the cytoplasmic face, and VirB4 associates with the inner membrane. VirB5 is a minor pilin involved in pilus function and possibly host cell interaction. VirB6 is a multi-pass inner membrane protein; VirB7 is a lipoprotein that anchors the complex in the outer membrane; VirB8 is an inner membrane component important for assembly; VirB9 and VirB10 contribute to the core channel spanning the periplasm and outer membrane.

Genetic organization and regulation: virB1 through virB11 are often contiguous on Ti plasmids and are induced as part of the vir gene response upon plant signal perception, primarily controlled by the VirA/VirG two-component regulatory system. The VirB apparatus works in concert with other virulence factors such as VirD2 and VirE2 that participate in T-DNA processing and protection during transfer, though these latter genes lie outside the virB cluster.

Significance: the virB1virB11 locus is central to the mechanism of DNA and protein transfer in conjugation-like systems and has provided key insights into secretion biology and biotechnological applications for DNA delivery.
