Ubiquitinmarkierte, often translated as ubiquitinated, refers to a protein that has been covalently tagged with one or more molecules of ubiquitin. Ubiquitin is a small, highly conserved protein found in almost all eukaryotic organisms. The process of attaching ubiquitin to a target protein is called ubiquitination, and it is a crucial post-translational modification that regulates a vast array of cellular processes.

Ubiquitination is a multi-step enzymatic cascade involving three classes of enzymes: E1 (ubiquitin-activating enzyme), E2 (ubiquitin-conjugating enzyme), and E3 (ubiquitin ligase). The E3 ligase plays a critical role in determining substrate specificity, recognizing and binding to the target protein. Once attached, ubiquitin can signal for various cellular fates.

A common consequence of ubiquitination is the targeting of the marked protein for degradation by the 26S proteasome, a cellular machine responsible for breaking down damaged or no longer needed proteins. However, ubiquitination can also have non-degradative functions. For example, it can alter protein localization, modulate protein activity, facilitate protein-protein interactions, or play a role in DNA repair and signal transduction pathways. The type and number of ubiquitin molecules attached, as well as the specific lysine residue on the ubiquitin chain, can dictate the resulting cellular outcome, creating a complex signaling code.
