Troponiinitropomysiinisäätely refers to the molecular mechanism that controls muscle contraction, specifically in striated muscle. This process involves the interaction of three key proteins: actin, myosin, and the troponin-tropomyosin complex. Actin and myosin are the contractile proteins, forming the filaments that slide past each other to shorten the muscle. Tropomyosin is a long, fibrous protein that lies along the actin filament, and troponin is a complex of three smaller proteins (troponin C, troponin I, and troponin T) that is attached to tropomyosin.

In a relaxed muscle, tropomyosin is positioned in a way that physically blocks the binding sites on actin where myosin heads would normally attach. This blockage prevents the formation of cross-bridges between actin and myosin, and thus prevents contraction. The regulation of this blockage is achieved through the troponin complex.

When a muscle is stimulated by a nerve impulse, it triggers the release of calcium ions (Ca2+) from the sarcoplasmic reticulum into the sarcoplasm. Calcium ions then bind to troponin C, a component of the troponin complex. This binding causes a conformational change in the entire troponin complex. This conformational change, in turn, pulls tropomyosin away from the myosin-binding sites on actin. With the binding sites now exposed, myosin heads can bind to actin, initiating the cross-bridge cycle. This cycle involves the repeated binding, pulling, and release of myosin heads, leading to the sliding of actin filaments over myosin filaments and ultimately muscle contraction.

When the nerve stimulation stops and calcium ions are pumped back into the sarcoplasmic reticulum, the concentration of calcium ions in the sarcoplasm decreases. Calcium dissociates from troponin C, causing the troponin-tropomyosin complex to return to its original position, re-blocking the myosin-binding sites on actin and allowing the muscle to relax.
