Thioesterbindung

Thioesterbindung refers to a type of chemical bond that is structurally similar to an ester bond, but with a sulfur atom replacing the oxygen atom in the carbonyl group. The general formula for a thioester is R-C(=O)-S-R', where R and R' are organic groups. This functional group plays a crucial role in various biological and chemical processes.

In biochemistry, thioester bonds are important energy carriers, comparable to high-energy phosphate bonds in molecules like ATP. The cleavage of a thioester bond releases a significant amount of free energy, which can be harnessed to drive other biochemical reactions. Acetyl-CoA is a well-known example of a molecule containing a thioester bond, serving as a central intermediate in metabolism, particularly in the citric acid cycle and fatty acid synthesis. Other biological thioesters include succinyl-CoA and acyl carrier protein derivatives.

Chemically, thioesters are more reactive than their oxygen ester counterparts. The carbon-sulfur bond is weaker than the carbon-oxygen bond, making thioesters more susceptible to nucleophilic attack. This increased reactivity is exploited in certain organic synthesis reactions. Thioesters can be synthesized through various methods, often involving the reaction of a carboxylic acid with a thiol, typically in the presence of a coupling agent.
