Variation among isoforms, species, or environmental conditions (pH, temperature) can further modulate specificity.

Studying substrate specificity involves kinetic assays, structural biology techniques (X‑ray crystallography, NMR), and computational docking. Engineered alterations—through directed evolution or rational design—aim to expand, shift, or refine specificity for applications in medicine, biotechnology, and industrial catalysis. Substrate specificity thus shapes metabolic pathways, drug interactions, and the evolution of enzymatic function.
