Protonation and deprotonation describe the gain or loss of a proton (hydrogen ion, H+) by a chemical species. Protonation is the process by which a molecule or ion accepts a proton, often increasing its positive charge or altering its basicity. Deprotonation is the reverse process, in which a molecule loses a proton, forming a conjugate base and usually becoming less positively charged. These processes are central to acid-base chemistry and operate in many solvents, most importantly water.

Under Brønsted-Lowry theory, acids are proton donors and bases are proton acceptors. The extent of protonation or deprotonation depends on pH and the intrinsic acidity or basicity of the site, often expressed as pKa. When the pH is below the pKa, the protonated form predominates; when the pH is above the pKa, the deprotonated form dominates. The Henderson-Hasselbalch equation relates pH, pKa, and the ratio of the two forms.

Common examples illustrate these ideas: ammonia is protonated to ammonium (NH4+); acetic acid loses a proton to form acetate (CH3COO−). Amino acids possess multiple ionizable groups that shift charge states with pH, influencing structure and function. Water autoionizes to H3O+ and OH−, a small but essential protonation-deprotonation equilibrium in aqueous solutions.

Protonation states affect reactivity, solubility, binding, and enzymatic catalysis, making pKa values important in chemistry and biology. In practice, protonation and deprotonation are probed by pH titrations, NMR chemical shifts, and other spectroscopic methods, and can occur rapidly on experimental timescales. Understanding these processes aids in drug design, catalysis, and the behavior of biomolecules.
