Proteiinimääritykset refer to the methods used to quantify the amount of protein present in a sample. These techniques are fundamental in various biological and chemical research fields, as protein concentration is a critical parameter for understanding cellular processes, enzyme activity, and drug development.

Common methods for protein quantification rely on either direct measurement of protein properties or indirect estimation based on the presence of specific protein components. Spectrophotometric methods are widely used, with the Bradford assay and the bicinchoninic acid (BCA) assay being popular choices. The Bradford assay utilizes Coomassie Brilliant Blue dye, which binds to proteins and causes a color change proportional to protein concentration. The BCA assay also relies on a colorimetric reaction, but it specifically detects peptide bonds and certain amino acid side chains. Both methods are relatively quick and easy to perform but can be affected by the presence of interfering substances in the sample.

Another approach involves measuring the absorbance of ultraviolet (UV) light by proteins, typically at a wavelength of 280 nm. This absorbance is due to the presence of aromatic amino acids like tryptophan and tyrosine. While straightforward, this method is less sensitive and can be inaccurate if the protein composition varies significantly or if other UV-absorbing compounds are present.

For more precise and specific protein quantification, techniques like enzyme-linked immunosorbent assays (ELISAs) can be employed. ELISAs use antibodies to specifically bind to a target protein, allowing for highly sensitive and quantitative measurement. Other methods include quantitative amino acid analysis, mass spectrometry, and SDS-PAGE coupled with densitometry, each offering different advantages in terms of specificity, sensitivity, and sample compatibility. The choice of protein quantification method depends on factors such as the sample matrix, the required sensitivity and accuracy, and the presence of potential interfering substances.
