Proteasomisetärjestelmän, commonly known as the ubiquitin-proteasome system (UPS), is a crucial cellular mechanism responsible for the targeted degradation of proteins. This complex pathway plays a vital role in maintaining cellular homeostasis, regulating a wide array of biological processes, and eliminating damaged or misfolded proteins. The UPS is comprised of two main components: the ubiquitin tagging system and the proteasome itself.

Ubiquitination is a post-translational modification where a small protein called ubiquitin is covalently attached to a target protein. This process is carried out by a cascade of enzymes, including ubiquitin-activating enzymes (E1), ubiquitin-conjugating enzymes (E2), and ubiquitin ligases (E3). E3 ligases are particularly important as they confer substrate specificity, determining which proteins will be targeted for degradation. The attachment of a polyubiquitin chain to a protein signals it for recognition by the proteasome.

The proteasome is a large, multi-subunit protein complex that acts as the cellular garbage disposal. It unfolds the ubiquitinated protein and then cleaves it into small peptides, which are subsequently broken down into individual amino acids by other cellular peptidases. The UPS is essential for processes such as cell cycle progression, DNA repair, immune response, and the removal of misfolded proteins that could otherwise accumulate and cause cellular dysfunction or disease. Dysregulation of the UPS has been implicated in various pathological conditions, including cancer, neurodegenerative diseases, and autoimmune disorders, making it a significant area of research.
