Promotormethylatie refers to the process where a methyl group is added to a DNA molecule. Specifically, this modification often occurs at cytosine bases that are followed by a guanine base, forming what are known as CpG sites. These CpG sites are frequently clustered in regions called CpG islands, which are commonly found in the promoter regions of genes. The promoter is a critical DNA sequence that regulates the transcription of a gene.

The addition of a methyl group to DNA, a process known as DNA methylation, is a fundamental epigenetic mechanism. Epigenetic modifications do not alter the underlying DNA sequence but can significantly influence gene expression. In the context of promoter regions, methylation typically leads to gene silencing.
