Promoterkontekst

Promoterkontekst is a term used in genetics and genomics to describe the sequence and chromatin environment surrounding a gene promoter that influences transcription initiation and regulation. It includes the core promoter region, which contains elements such as the TATA box, initiator (Inr), and downstream promoter element (DPE) that help recruit RNA polymerase II, as well as proximal promoter elements like the CAAT box and GC box that modulate promoter strength. The promoter context also encompasses distal regulatory elements, including enhancers, silencers, and insulators, and the three-dimensional folding of chromatin which brings these elements into physical contact with the promoter through looping.

The local chromatin landscape—nucleosome positioning, DNA methylation, and histone modifications such as H3K4me3 at active promoters and H3K27ac—shapes promoter accessibility and activity. In addition, transcription factor binding and cofactor recruitment within the promoter neighborhood contribute to tissue-specific and developmental regulation, and can influence promoter directionality and responsiveness to signaling.

Methods to study promoter context include reporter assays and massively parallel reporter assays (MPRA) to quantify promoter activity, as well as genomic approaches such as ATAC-seq and ChIP-seq to map chromatin state and factor occupancy, and chromosome conformation capture techniques (3C, Hi-C) to detect promoter–enhancer interactions.
