Peptidoglycans

Peptidoglycans are essential structural components of bacterial cell walls, providing mechanical strength and shape. They are complex, mesh-like polymers composed of sugar and amino acid building blocks. The primary sugar components are N-acetylglucosamine (NAG) and N-acetylmuramic acid (NAM), which are linked by β-(1,4)-glycosidic bonds. These chains are cross-linked by short peptides, or amino acid chains, forming a lattice that enwraps the bacterial cell.

The peptidoglycan layer is critical for maintaining cellular integrity, especially in hypotonic environments where osmotic pressure can cause cell lysis. Its sturdy but dynamic network allows bacteria to grow and divide by remodeling the structure through enzymatic activities involved in synthesis and degradation.

Peptidoglycans vary significantly among bacterial species, influencing cell shape and structural properties. For instance, gram-positive bacteria possess a thick peptidoglycan layer, which can be several layers thick, while gram-negative bacteria have a thinner layer situated between the inner cell membrane and an outer membrane.

Because of its unique composition, peptidoglycan is an important target for certain antibiotics, such as penicillin, which inhibits enzymes involved in peptidoglycan cross-linking, thereby weakening the bacterial cell wall and leading to cell death. Additionally, peptidoglycan fragments can act as pathogen-associated molecular patterns (PAMPs), triggering immune responses in hosts.
