PFKFB2

PFKFB2, also known as 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 2, is an enzyme that plays a crucial role in glucose metabolism. It is a bifunctional enzyme, meaning it possesses two distinct enzymatic activities. The first activity is phosphofructokinase, which catalyzes the synthesis of fructose-2,6-bisphosphate from fructose-6-phosphate. The second activity is fructose-2,6-bisphosphatase, which catalyzes the hydrolysis of fructose-2,6-bisphosphate to fructose-6-phosphate.

Fructose-2,6-bisphosphate is a key allosteric regulator of glycolysis, a metabolic pathway that breaks down glucose to produce energy. It activates phosphofructokinase-1 (PFK1), a rate-limiting enzyme in glycolysis, thereby promoting glucose breakdown. Conversely, it inhibits fructose-1,6-bisphosphatase, an enzyme involved in gluconeogenesis, the synthesis of glucose.

The expression and activity of PFKFB2 are regulated by various factors, including hormones like insulin and glucagon, as well as cellular energy status. In conditions of high glucose availability and insulin signaling, PFKFB2 activity increases, leading to higher levels of fructose-2,6-bisphosphate and enhanced glycolysis. In contrast, during fasting or low energy states, PFKFB2 activity is reduced, favoring gluconeogenesis.

PFKFB2 is found in various tissues, with particularly high expression in the heart and skeletal muscle. Its role in energy metabolism makes it important for maintaining cellular energy homeostasis. Dysregulation of PFKFB2 has been implicated in metabolic disorders such as diabetes and cancer, where altered glucose utilization is a hallmark.
