Nitrophenyl

Nitrophenyl refers to a substituted phenyl group bearing a nitro group (NO2). In organic chemistry the term is used to designate any phenyl ring in which one hydrogen is replaced by a nitro group. The nitro substituent is typically located at the ortho, meta, or para position, with o-nitrophenyl, m-nitrophenyl, and p-nitrophenyl variants being common. The position of substitution influences reactivity and properties.

The nitro group is strongly electron withdrawing, deactivating the ring toward electrophilic substitution and altering the acidity and UV-visible spectra of the compound. p-Nitrophenyl derivatives are especially notable because the p-nitrophenyl moiety itself is yellow and serves as a chromogenic leaving group in various assays. p-Nitrophenyl esters and carbonates are activated esters used in bioconjugation and peptide synthesis; hydrolysis of p-nitrophenyl esters releases p-nitrophenol, which under basic conditions turns yellow and absorbs at around 405 nm, enabling spectrophotometric detection.

Preparation and applications: Nitrophenyl groups are typically introduced by electrophilic nitration of appropriate precursors or by substitution of halogenated phenyl derivatives. In practical use, nitrophenyl esters such as p-nitrophenyl acetate or p-nitrophenyl chloroformate function as activated reagents in organic synthesis and biochemical assays. In dye chemistry and materials science, nitrophenyl motifs occur in nitroaromatic dyes and polymers.
