NMethylDaspartate

N-methyl-D-aspartate (NMDA) is a synthetic derivative of the amino acid aspartic acid, existing as the D-enantiomer. It is a potent, selective agonist for NMDA-type glutamate receptors in the central nervous system and is widely used in neuroscience research to activate these receptors and study excitatory neurotransmission, synaptic plasticity, and calcium signaling pathways.

NMDA receptors are ligand-gated ion channels that require binding of glutamate and a co-agonist (glycine or D-serine) for activation. NMDA binds to its receptor site, and channel opening depends on membrane depolarization to relieve a magnesium block. This allows influx of calcium and sodium (and efflux of potassium), triggering downstream signaling. The resulting calcium signaling contributes to processes such as long-term potentiation and learning and memory. However, excessive NMDA receptor activation can cause calcium overload and excitotoxic neuronal injury, a mechanism implicated in stroke and other neurological conditions.

In research, NMDA is used to elicit strong NMDA receptor–mediated responses in brain slices, neuronal cultures, and animal models. It helps delineate receptor pharmacology, synaptic development, and neurodegenerative mechanisms. Clinically, NMDA itself is not used as a therapeutic agent due to toxicity; by contrast, NMDA receptor antagonists (for example memantine or ketamine) have therapeutic applications in certain contexts.
