Mutagenitas

Mutagenitas is a term used in genetics and toxicology to denote the mutagenic potential of a substance, agent, or exposure that can induce genetic mutations in DNA. The concept encompasses chemical, physical, and biological mutagens and is distinct from general toxicity or cytotoxicity. Etymology traces to Latin mutare (to change) and genetitas, reflecting the idea of inheritable changes.

Definition and scope: Mutagenitas describes the tendency of an agent to cause heritable DNA alterations, including base substitutions, insertions and deletions, and chromosomal rearrangements. The degree of mutagenicity depends on concentration, exposure duration, metabolic activation, and the biological system examined.

Mechanisms: Mutagenic effects arise through direct DNA damage, formation of DNA adducts, replication errors, or impaired DNA repair. Some agents require metabolic activation to become mutagenic. Oxidative stress and interference with replication can also contribute to mutagenesis.

Assessment: Mutagenitas is commonly evaluated with genotoxicity tests such as the Ames test, mammalian cell mutation assays, and assays detecting chromosomal damage like the micronucleus test or the comet assay. Results are usually expressed as a mutagenicity index, a fold increase in mutation rate, or a positive/negative outcome, and used in risk assessment.
