Hemateinmordant

Hemateinmordant refers to a histological staining system in which hematoxylin is oxidized to hematein and combined with a mordant metal to form a dye-lake that binds to tissue, producing nuclear staining. In routine practice, hematoxylin solutions are prepared or oxidized to hematein in the presence of a mordant such as aluminum or iron salts, enabling the dye to form an insoluble complex that adheres to basophilic structures like nucleic acids in cell nuclei.

The staining mechanism relies on the formation of a hematein–mordant complex that binds to negatively charged tissue components. The resulting nuclear color is typically blue to blue-black, with shade varying by mordant type, oxidation method, and counterstain. Blueing steps in alkaline conditions may be used after differentiation to enhance the blue nuclear appearance. The process underpins many common stains in histology, especially hematoxylin and eosin (H&E) protocols, where hematein–mordant complexes provide the nuclear contrast essential for cellular morphology.

Common mordants include aluminum salts and iron salts, and several commercial formulations exist, each yielding slightly different staining characteristics. Variants of the approach are designed to optimize tissue penetration, staining intensity, and differentiation. While the term is frequently encountered in older literature and in language variants of hematoxylin protocols, the practical effect remains the same: a metal mordant enables the oxidized hematoxylin to form a stable, tissue-bound complex that highlights nuclei.
