GolginTGNalueelta

GolginTGNalueelta is a member of the Golgin family of coiled-coil tethering proteins that localizes to the trans-Golgi network (TGN). Golgins are large cytosolic scaffolds that extend from the Golgi membrane to capture incoming vesicles and organize the Golgi apparatus. GolginTGNalueelta is proposed to act as a long tether that helps dock vesicles arriving at the TGN and to contribute to cargo sorting.

Localization and structure: It concentrates at the TGN in mammalian cells, often detected as perinuclear Golgi-associated signal overlapping markers such as TGN46. The protein is predicted to contain extensive coiled-coil domains that form elongated dimers or oligomers, enabling a tethering scaffold, and a membrane-anchoring region near the C‑terminus.

Function: By capturing transport vesicles and presenting them to SNARE complexes, GolginTGNalueelta facilitates fusion with the TGN membrane and ensures efficient delivery of secretory and lysosomal enzymes. Interactions: GolginTGNalueelta is thought to interact with small GTPases of the Rab family, including Rab6 and Rab33b, as well as SNARE proteins involved in TGN fusion. It likely acts in concert with other Golgin tethers to coordinate vesicle capture and membrane remodeling.

Regulation and context: Expression of GolginTGNalueelta is widespread in mammalian tissues; activity may be regulated through cell cycle-dependent phosphorylation, and signaling pathways controlling Golgi dynamics. Functional perturbation through RNAi or CRISPR can lead to delayed trafficking, altered Golgi morphology, or mis-sorting of cargos in cell-based assays.
