GABAreceptormodulers are a class of chemical compounds that interact with gamma-aminobutyric acid (GABA) receptors in the central nervous system. GABA is the primary inhibitory neurotransmitter in the brain, playing a crucial role in regulating neuronal excitability. Modulators can either enhance or reduce the activity of these receptors.

Positive allosteric modulators (PAMs) bind to a site on the GABA receptor distinct from the GABA binding site, increasing the receptor's affinity for GABA or its channel opening probability. This leads to a greater inhibitory effect. Benzodiazepines, for example, are well-known PAMs of the GABAA receptor, used to treat anxiety, insomnia, and seizures. Negative allosteric modulators (NAMs) have the opposite effect, decreasing the receptor's response to GABA. This can lead to increased neuronal excitability. While less common therapeutically, some NAMs have been investigated for their potential in treating conditions like epilepsy where excessive inhibition is problematic.

Other types of modulators exist, including competitive antagonists that block GABA from binding to its receptor, or non-competitive antagonists that bind within the ion channel pore, preventing ion flow. The precise interaction and resulting effect depend on the specific subtype of GABA receptor and the structure of the modulator.
