Functionalgrouptolerant is a term used in chemistry and related fields to describe a system, typically a chemical reaction, catalyst, or process, that can operate effectively in the presence of a wide range of functional groups on its substrates. A functionalgrouptolerant method minimizes the need for protective groups and redox or functional-group manipulation, enabling broader substrate scope and streamlined workflows.

In synthetic organic chemistry, high functional-group tolerance is a key goal for methods such as cross-coupling, oxidation, and C–H functionalization. Reagents or catalysts described as functionalgrouptolerant are expected to withstand common functional groups—such as esters, amides, nitriles, nitro groups, halides, alcohols, and ketones—without undergoing side reactions that disable the transformation. Nonetheless, tolerance is not universal; certain groups can poison catalysts, promote side reactions, or require specific conditions to be tolerated.

Tolerance is typically assessed by substrate scope studies. Researchers compile tolerance maps or tables showing yields and selectivities across substrates bearing diverse functional groups. Factors that enhance tolerance include catalyst design (ligand environment, metal choice), solvent and additives, temperature, and reaction time. Trade-offs are common: expanding tolerance for one class of groups may reduce reactivity or selectivity for others.

Beyond synthesis, the concept also appears in biocatalysis and materials science, where compatibility with various functional groups affects enzyme activity or material performance.
