In scientific discussions, the best-known form of this concept is FOXO4-DRI, which has been investigated as a potential senolytic strategy to target senescent cells.

Mechanism and purpose: In many aging-related models, senescent cells rely on a FOXO4–p53 interaction to maintain survival and nuclear retention of p53. FOXO4-DRI is designed to bind FOXO4 and block its interaction with p53, freeing p53 from the nucleus and promoting apoptosis of senescent cells. The peptide is rendered proteolytically stable by using a D-enantiomer configuration, which can improve its half-life in biological systems and support systemic delivery in preclinical settings.
