Enzyme activity is typically measured in units, often defined as the amount of substrate converted to product per unit of time, or the amount of product formed per unit of time.

Factors affecting enzyme activity can be understood through specific principles. Substrate concentration generally increases reaction rate until the enzyme becomes saturated, meaning all active sites are occupied. Temperature also influences activity, with an optimal temperature range where activity is highest. Beyond this optimum, increasing temperature can lead to denaturation, a process where the enzyme loses its functional three-dimensional structure and thus its activity. Similarly, pH affects enzyme activity. Each enzyme has an optimal pH range, and deviations from this optimum can alter the ionization state of amino acid residues in the active site or the enzyme's overall structure, leading to reduced activity.

Activators are substances that increase enzyme activity, often by binding to the enzyme and enhancing its catalytic efficiency or by facilitating substrate binding. Inhibitors, conversely, decrease enzyme activity. They can bind to the enzyme in reversible or irreversible ways, interfering with the enzyme's ability to bind substrate or perform catalysis.
