Endsubstrat

Endsubstrat is a term used in biochemistry and enzymology to denote the final substrate in a sequential enzymatic process or the substrate that is acted upon before the formation of the end product. The concept is employed to describe substrates within multi-step cascades where distinct enzymes act in sequence, helping to clarify the order of substrates and reactions.

The name combines the idea of an endpoint (end) with the conventional notion of a substrate. Its usage is not universally standardized; some authors treat endsubstrat as synonymous with the terminal substrate or the last substrate before product formation, while others reserve it for a substrate that becomes limiting for the final step in a pathway. In kinetic modeling, endsubstrat may be considered the substrate whose depletion most directly constrains the rate of the final reaction, or a substrate that accumulates if the downstream enzyme is slow or saturated.

In practice, endsubstrat can be quantified alongside intermediates and products using methods such as chromatography or mass spectrometry, enabling researchers to track substrate flux through a pathway. The term is often contrasted with starting substrate, which enters the pathway, and with intermediates, which appear transiently en route to the endsubstrate and final product.

Applications of the concept appear in metabolic engineering and systems biology, where clarifying the sequence of substrates can aid pathway optimization and the interpretation of kinetic data. Some authors prefer terms like terminal substrate or final substrate to avoid ambiguity.
