Endoteelinjärjestelmä refers to the system of cells and molecules involved in the production and action of endothelins. Endothelin is a potent vasoconstrictor peptide, meaning it causes blood vessels to narrow. There are three main types of endothelin: ET-1, ET-2, and ET-3. ET-1 is the most abundant and physiologically important. Endothelins are primarily synthesized and released by endothelial cells, which form the inner lining of blood vessels. However, they can also be produced by other cell types, such as smooth muscle cells and certain neurons.

The synthesis of endothelin-1 is a complex process involving several steps. It begins with the transcription of the endothelin gene, followed by translation into a precursor protein. This precursor is then processed through several intermediate stages before the mature, active endothelins are formed and released. Endothelin's effects are mediated by binding to specific receptors, primarily ETA and ETB receptors, which are found on various cell types, including vascular smooth muscle cells, endothelial cells, and kidney cells. Binding to ETA receptors typically leads to vasoconstriction, while binding to ETB receptors can cause either vasodilation or vasoconstriction depending on the cell type and context.

The endothelins play crucial roles in regulating blood pressure, vascular tone, and cell growth. Dysregulation of the endothelins has been implicated in a variety of cardiovascular and pulmonary diseases, including hypertension, pulmonary hypertension, atherosclerosis, and heart failure. Consequently, the endothelin system has become a significant target for therapeutic intervention, with endothelin receptor antagonists being developed and used to treat certain conditions.
