The Endoplasmic Reticulum Membrane Model, often referred to as the EndoplasmaMembranmodellen in German, describes the structure and function of the endoplasmic reticulum (ER) and its surrounding membrane. The ER is a complex network of membranes found throughout the cytoplasm of eukaryotic cells. It exists in two forms: the rough endoplasmic reticulum (RER), studded with ribosomes, and the smooth endoplasmic reticulum (SER), lacking ribosomes.

The membrane of the ER is a lipid bilayer composed primarily of phospholipids and proteins, similar to the plasma membrane but with distinct protein compositions that dictate its specific functions. This membrane encloses an internal space called the lumen. The RER membrane is involved in protein synthesis, folding, modification, and transport, particularly for proteins destined for secretion or insertion into other membranes. Ribosomes attached to the RER surface synthesize these proteins and translocate them into the ER lumen or embed them within the ER membrane.

The SER membrane plays a crucial role in lipid synthesis, steroid hormone production, detoxification of drugs and poisons, and calcium storage. The fluidity of the ER membrane allows for dynamic changes in its structure, including the formation of vesicles that bud off from the ER to transport molecules to other organelles, such as the Golgi apparatus. The continuity of the ER membrane with the outer nuclear envelope further highlights its integrated role within the cell's endomembrane system.
