EOMES

Eomes, short for eomesodermin (gene symbol EOMES), is a transcription factor in the T-box (TBX) family. The protein contains a conserved T-box DNA-binding domain and regulates gene expression programs that direct cell fate during development and immune cell differentiation.

Expression patterns of EOMES vary across development and cell types. In vertebrate embryos, it is expressed in the early trophectoderm and other extraembryonic lineages, and later in developing neural and hematopoietic tissues. In the immune system, EOMES is expressed by natural killer (NK) cells and by subsets of CD8+ T cells, as well as by certain innate-like T cells such as iNKT cells.

Developmental roles are prominent in mouse models. Eomes contributes to placental development and trophoblast lineage formation, supporting proper mesoderm differentiation and extraembryonic functions. Loss of Eomes can result in embryonic abnormalities and placentation defects, underscoring its essential role in early development.

In the immune system, EOMES drives cytotoxic differentiation and effector function. It promotes expression of granzyme B, perforin, and IFN-γ, supports the development of memory CD8+ T cells, and influences NK cell maturation. EOMES operates with other transcription factors such as TBX21 (T-bet) and RUNX3 to shape lineage outcomes, with cytokine signals like IL-15 and IL-2 capable of inducing its expression.

Clinical and research relevance includes investigations into cancer immunology and responses to viral infections, where EOMES helps determine the strength and quality of cytotoxic responses. In humans, germline EOMES mutations are rare, and most insights come from cellular models and conditional mouse studies.
