DMC1, or DNA meiosis 1 recombinase, is a conserved meiosis-specific recombinase found in a wide range of eukaryotes. It is a homolog of the bacterial RecA protein and plays a central role in homologous recombination during meiosis. In germ cells, DMC1 forms nucleoprotein filaments on single-stranded DNA and promotes the pairing and strand exchange between homologous chromosomes, a process that leads to crossovers necessary for accurate chromosome segregation.

The activity of DMC1 is ATP-dependent and relies on the assembly of filamentous structures that search for homology and catalyze strand invasion. In many organisms, DMC1 works in concert with RAD51 and a set of accessory factors, including the HOP2–MND1 heterodimer, which stabilize filaments and stimulate recombination. This cooperation helps ensure interhomolog recombination rather than intersister repair, contributing to genetic diversity and proper chromosomal alignment during meiosis.

Expression of DMC1 is typically restricted to germ cells undergoing meiosis, and the protein localizes to chromosome axes and the synaptonemal complex. Its activity is tightly regulated to prevent inappropriate recombination in somatic cells.

Mutations or functional defects in DMC1 disrupt meiotic recombination, leading to reduced fertility, defective chromosome pairing, and increased risk of chromosomal missegregation and aneuploidy in several species. DMC1 remains a key focus in studies of meiotic biology, fertility, and genome stability, providing insight into the mechanisms that ensure accurate genetic inheritance.
