Diszulfidkötés refers to a covalent chemical bond formed between two sulfur atoms, typically in organic molecules. This bond is often denoted as -S-S-. It is particularly important in biochemistry, where it plays a crucial role in the structure and function of many proteins. These disulfide bridges, as they are commonly called in biological contexts, can form between cysteine residues within a single polypeptide chain, contributing to its tertiary structure, or between different polypeptide chains, stabilizing quaternary structures.

The formation of a disulfide bond is an oxidation reaction, where two thiol (-SH) groups are oxidized to form the S-S linkage, releasing two hydrogen atoms. This process can be reversible, with disulfide bonds being reduced back to thiol groups. This reversible nature is essential for many biological processes, allowing proteins to undergo conformational changes. Enzymes like protein disulfide isomerase (PDI) facilitate the formation and rearrangement of disulfide bonds within proteins, ensuring they adopt their correct functional shapes. Disruptions in disulfide bond formation or maintenance can lead to protein misfolding and associated diseases. Disulfide bonds are also found in other molecules beyond proteins, contributing to their chemical properties.
