Corepressorabhängige (CR) are a class of proteins that play a crucial role in the regulation of gene expression in eukaryotic cells. They are characterized by their ability to interact with and modulate the activity of transcription factors, which are proteins that bind to specific DNA sequences to regulate the transcription of genes. Corepressors are typically associated with the repression of gene expression, acting as negative regulators of transcription.

Corepressors can be classified into several families based on their structural and functional characteristics. One of the most well-studied families is the SMRT/NCoR family, which includes proteins like SMRT (Silencing Mediator of Retinoic Acid and Thyroid hormone receptor) and NCoR (Nuclear Receptor Corepressor). These proteins contain multiple conserved domains, including a nuclear receptor interaction domain (NID), a ligand-independent activation domain (LID), and a transcriptional repression domain (TRD). The NID and LID domains mediate the interaction with transcription factors, while the TRD interacts with the basal transcription machinery to repress gene expression.

Another family of corepressors is the Groucho/TLE family, which includes proteins like Groucho and TLE (Transducin-like enhancer of split). These proteins contain a conserved Groucho domain (GCD) that mediates their interaction with transcription factors. The GCD is also found in other proteins, such as the zinc finger proteins and the homeobox proteins, suggesting that it may have evolved from a common ancestral gene.

Corepressors can also be classified based on their mechanism of action. Some corepressors, like SMRT and NCoR, act as direct inhibitors of transcription by interacting with the basal transcription machinery. Others, like Groucho and TLE, act as indirect inhibitors by competing with coactivators for binding to transcription factors.

The study of corepressors has important implications for our understanding of gene regulation and its role in various biological processes, including development, differentiation, and disease. By modulating the activity of transcription factors, corepressors can fine-tune gene expression in response to environmental cues and cellular signals, ensuring that the appropriate genes are expressed at the appropriate time and in the appropriate cell type.
