Copperresponsive refers to the property of biological systems, organisms, and engineered circuits that detect and respond to copper ion concentrations. These responses help maintain copper homeostasis, balancing copper's essential catalytic roles with its high reactivity and potential toxicity.

In bacteria and archaea, copper sensing is mediated by metalloregulatory proteins such as CueR, CsoR, and CopY. These regulators bind copper and then modulate transcription of operons involved in copper uptake, storage, detoxification, and efflux. Common components include copper transporters (like P-type ATPases), copper chaperones that safely deliver copper to specific enzymes, and efflux systems that reduce cytosolic copper.

In eukaryotes, copper-responsive control involves metallochaperones, metallothioneins, and copper-transporting ATPases (for example ATP7A and ATP7B in humans), along with transcriptional networks that adjust expression of copper-dependent enzymes and storage proteins in response to copper status.

Copperresponsive elements and promoters are exploited in research and biotechnology to create biosensors and programmable circuits that report environmental copper levels or regulate gene expression in response to copper. Disruption of copper homeostasis is linked to human diseases such as Wilson disease and Menkes disease, reflecting the tight regulation required for this metal.
