COL19A1

COL19A1 is a protein-coding gene in humans that encodes the collagen alpha‑1(XIX) chain, the main component of collagen type XIX. The encoded protein belongs to the collagen superfamily but differs from the typical fibrillar collagens because it has a short N‑terminal non‑collagenous domain, a central triple‑helical Gly‑X‑Y repeat region, and a C‑terminal non‑collagenous domain. Collagen type XIX is a non‑fibrillar collagen that is expressed predominantly in cartilage, bone, spinal cord, and various brain regions, suggesting a role in the extracellular matrix and in tissue development.

The COL19A1 gene is located on chromosome 12 at band q13.12. The gene undergoes alternative splicing, producing several transcript variants that encode proteins differing mainly at their non‑collagenous termini. Transcription of COL19A1 can be regulated by transforming growth factor‑β (TGF‑β) signaling, and its expression is increased during chondrogenic differentiation of mesenchymal stem cells.

Functional studies in animal models indicate that collagen XIX contributes to the structural integrity of cartilage and may influence the regulation of cell proliferation and differentiation in developing bone and cartilage. Although mutations in COL19A1 are not commonly associated with human disease, alterations in its expression levels have been observed in conditions involving cartilage degeneration, such as osteoarthritis, and in some developmental skeletal disorders.
