12SHPETE

12S-hydroperoxyeicosatetraenoic acid (12S-HPETE) is a hydroperoxide derivative of arachidonic acid produced in the lipoxygenase pathway. It is one of several hydroperoxyeicosatetraenoic acids formed when molecular oxygen is added to arachidonic acid, with a hydroperoxy group at carbon 12 and the S configuration at that center.

Formation and metabolism: In humans, 12-lipoxygenase (12-LOX) catalyzes the insertion of oxygen at C12 of arachidonic acid to yield 12S-HPETE. This unstable intermediate is typically rapidly reduced by cellular peroxidases to the corresponding 12S-hydroxy metabolite (12S-HETE). 12S-HPETE can also be diverted into additional enzymatic pathways, contributing to the broader network of eicosanoid metabolism that generates diverse signaling molecules.

Biological role: 12S-HPETE acts as a signaling lipid involved in inflammatory responses. While its direct activities are less extensively characterized than those of its downstream products, it is considered a precursor or upstream mediator in pathways that regulate leukocyte function, vascular tone, and other aspects of inflammation. Its levels and flux through LOX-related pathways can influence the production of various eicosanoids and related mediators, with potential relevance to conditions such as inflammatory diseases and cancer.
