1,2-Diglycerides, also known as 1,2-diacylglycerols or abbreviated as DAGs, are a class of lipids that are esters of glycerol with two fatty acids. Specifically, the fatty acids are attached to the sn-1 and sn-2 positions of the glycerol backbone. This distinguishes them from 1,3-diglycerides, where the fatty acids are attached to the sn-1 and sn-3 positions. 1,2-Diglycerides are important intermediates in lipid metabolism and signaling pathways.

In biological systems, 1,2-diglycerides are primarily formed through the hydrolysis of phospholipids, a process often catalyzed by enzymes like phospholipase C. They also arise from the breakdown of triglycerides. Once formed, 1,2-diglycerides can serve as substrates for the synthesis of other lipids, such as phospholipids and triglycerides.

Beyond their role in lipid synthesis and breakdown, 1,2-diglycerides are significant signaling molecules. They act as second messengers in various cellular pathways, particularly those involving protein kinase C (PKC). Upon activation by certain stimuli, PKC translocates to the cell membrane and binds to DAG, leading to the phosphorylation of target proteins and subsequent cellular responses. The specific fatty acid composition of the 1,2-diglyceride can influence its signaling activity. They are found in various food sources and are also utilized as emulsifiers in the food industry.
