1,2-Diglyceride refers to a class of lipids composed of a glycerol backbone with two fatty acid molecules esterified to the first and second carbon atoms of the glycerol. The third carbon atom remains esterified to a hydrogen atom or is free. This distinguishes it from 1,3-diglycerides, where fatty acids are attached to the first and third carbons, and triglycerides, where all three hydroxyl groups of glycerol are esterified with fatty acids.

The presence of a free hydroxyl group on the third carbon of the glycerol backbone in 1,2-diglycerides makes them more polar than triglycerides. This amphipathic nature, with both hydrophilic (the hydroxyl group) and lipophilic (the fatty acid chains) regions, allows them to interact with both water and lipids. Consequently, diglycerides can act as emulsifiers, helping to stabilize mixtures of oil and water.

1,2-Diglycerides can be formed through the hydrolysis of triglycerides, a process that occurs during digestion or through enzymatic action. They are also intermediates in the synthesis of other lipids, including phospholipids and triglycerides. In biological systems, 1,2-diglycerides play roles in cell signaling pathways, particularly as activators of protein kinase C, an enzyme involved in a wide range of cellular processes.

While less common as dietary fats compared to triglycerides, diglycerides can be found in small amounts in various foods, especially those containing fats and oils. Their emulsifying properties are also utilized in the food industry as food additives to improve texture and stability in products like margarine and baked goods.
