Tropokollageenimolekyylejä are the soluble precursors to collagen fibrils, the primary structural proteins found in connective tissues of animals. These molecules are synthesized intracellularly as larger polypeptide chains, known as procollagen. Procollagen undergoes a series of post-translational modifications within the cell, including hydroxylation of proline and lysine residues, and glycosylation. Following these modifications, procollagen is secreted from the cell, where enzymes cleave off terminal propeptides, converting it into tropokollageenimolekyylejä.

Tropokollageenimolekyylejä are elongated, rod-like structures approximately 300 nm in length and 1.5 nm in diameter. They are composed of three polypeptide chains, called alpha chains, wound around each other in a triple helix. Each alpha chain typically contains about 1000 amino acids, with a repeating Gly-X-Y sequence where Gly is glycine, X is often proline, and Y is often hydroxyproline. This helical structure provides significant tensile strength to the resulting collagen.

Once secreted, tropokollageenimolekyylejä self-assemble spontaneously into a staggered array, forming quarter-staggered arrays. This arrangement creates overlap regions and gaps between the molecules. Subsequently, covalent cross-links form between adjacent tropokollageenimolekyylejä, further stabilizing the structure and leading to the formation of mature, insoluble collagen fibrils. These fibrils then aggregate into larger collagen fibers, which are essential for the structural integrity of tissues such as skin, bone, tendons, and cartilage.
