Trimannosyl is a biochemical descriptor used to denote oligosaccharide structures that contain three mannose residues. In glycoscience, trimannosyl motifs are a common feature of certain glycans, most notably the trimannosyl core of N-linked glycans, often represented as Man3GlcNAc2.

In the context of N-glycosylation, a core oligosaccharide is transferred to a nascent protein at an asparagine residue and includes three mannose units as part of its central core. After transfer, the core undergoes trimming and remodeling by specific mannosidases and glycosyltransferases, leading to a variety of trimannosyl-containing structures that can be further elaborated or partially trimmed to yield distinct glycan forms.

Trimannosyl motifs can appear in both high-mannose and paucimannose glycan families depending on the processing steps in the endoplasmic reticulum and Golgi apparatus. The term is used to describe this shared structural feature across different glycan contexts, rather than a single universal structure.

Functional and analytical relevance: The trimannosyl core plays a role in protein folding and quality control in the endoplasmic reticulum, influencing folding efficiency, trafficking, and stability. Recognition by lectins and glycan-binding proteins can be affected by the presence of a trimannosyl motif. In research and diagnostics, trimannosyl-containing glycans are analyzed using mass spectrometry, HPLC, and other glycomic techniques, and defined trimannosyl structures can be synthesized chemically or enzymatically for study or therapeutic development.
