Tetracycline-controlled refers to a set of inducible gene expression systems that use tetracycline or its analogs, such as doxycycline, to regulate transcription. These systems are widely used in molecular biology to achieve temporal and reversible control of gene activity in cells and organisms, including mammalian cell lines and transgenic animals. They were developed in the 1990s and have since become a standard tool for functional genomics and developmental studies.

At the core of tetracycline-controlled systems is a regulatory protein that responds to tetracycline. The two main variants are Tet-Off and Tet-On. Both rely on a tetracycline-responsive promoter containing Tet operator sequences (TetO) and a minimal core promoter. In the Tet-Off system, a tetracycline-controlled transcriptional activator (tTA) binds to TetO and activates transcription in the absence of tetracycline; when doxycycline is present, tetracycline binds tTA and prevents it from activating transcription, turning gene expression off. In the Tet-On system, a reverse regulator (rtTA) binds TetO and activates transcription only in the presence of doxycycline, turning gene expression on with inducer exposure.

Common components include the Tet-regulatory proteins (tTA or rtTA) and a Tet-responsive promoter driving the target gene. The system allows dose-dependent control and reversibility, enabling researchers to turn genes on or off by administering or withdrawing doxycycline. Applications span basic research, developmental biology, and disease modeling, with use in cultured cells and whole organisms. Considerations include potential leaky expression, the pharmacokinetics of doxycycline, and possible off-target effects, which can influence experimental design and interpretation.
