Sulfonamide-related refers to chemical entities that contain the sulfonamide group, a sulfonyl functional group bonded to an amino group. This structural motif appears in a broad range of compounds, including many medicines and some industrial chemicals. In pharmacology, sulfonamide-related drugs span antibiotic agents as well as non-antibiotic medications that rely on the same chemical core.

In medicine, the best-known sulfonamide-related drugs are the sulfonamide antibiotics, such as sulfamethoxazole and sulfadiazine. These agents act by inhibiting bacterial dihydropteroate synthase, an enzyme in the folate biosynthesis pathway, thereby blocking formation of essential nucleotides. They are frequently used in combination with trimethoprim, which inhibits a subsequent step in the same pathway, to yield a synergistic antibacterial effect. Resistance can arise through mutations in the target enzyme, increased PABA production, or reduced drug uptake.

Beyond antibiotics, sulfonamide-related compounds are found in various non-antibiotic drugs, including certain diuretics (for example, thiazide diuretics), carbonic anhydrase inhibitors (such as acetazolamide), and several sulfonylureas used to treat type 2 diabetes. The presence of the sulfonamide group can influence pharmacokinetic properties and the potential for hypersensitivity reactions.

Safety considerations are an important aspect of sulfonamide-related therapies. Patients may experience adverse effects ranging from mild skin reactions to more serious hypersensitivity. Nephrotoxicity and photosensitivity are possible, and clinicians consider cross-sensitivity with other sulfa-containing drugs when selecting therapy.
