Subcloned refers to a gene or DNA fragment that has been isolated from one DNA molecule and inserted into another, usually a plasmid or a different vector, for further manipulation or study. This process is a fundamental technique in molecular biology and genetic engineering. The original DNA molecule might be genomic DNA, a polymerase chain reaction (PCR) product, or another recombinant DNA molecule. The target DNA molecule, often called a cloning vector, typically contains elements that facilitate replication and selection, such as an origin of replication and an antibiotic resistance gene.

The purpose of subcloning can vary. It might be to express a gene in a specific host organism, to study the function of a particular DNA sequence by modifying it, or to generate a DNA probe for hybridization experiments. The process typically involves cutting both the source DNA and the vector with restriction enzymes, which create compatible ends. These ends are then joined together by DNA ligase, forming a new recombinant DNA molecule. The ligated product is then introduced into a host organism, usually bacteria like E. coli, through a process called transformation. Cells that have successfully taken up the recombinant DNA are then selected, often by plating them on a medium containing an antibiotic. Colonies that grow are then screened to confirm the presence and correct orientation of the subcloned DNA. This allows researchers to isolate and work with specific DNA segments efficiently.
