The measured signal is typically plotted on the y-axis, and the known concentration is plotted on the x-axis.

The fundamental principle behind a standard curve is that there is a reproducible relationship between the concentration of an analyte and the instrument's response. Once a sufficient number of data points are generated from standards of known concentrations, a mathematical model, often a linear regression line, is fitted to these points. This fitted line represents the calibration.

To determine the concentration of an unknown sample, the same analytical procedure used for the standards is applied to the unknown.
