The small subunit is the smaller component of a ribosome, the molecular machine responsible for translating genetic information into proteins. In the context of protein synthesis, the small subunit plays a central role in recognizing mRNA and ensuring correct decoding by tRNAs. It works in concert with the larger subunit to form a functional ribosome.

In bacteria, the small subunit is called the 30S, together with the large 50S subunit to form the 70S ribosome. In eukaryotes, the corresponding small subunit is the 40S, which combines with the large 60S subunit to yield the 80S ribosome. The small subunit contains ribosomal RNA and proteins; in prokaryotes, the characteristic rRNA is 16S rRNA, while in eukaryotes it is 18S rRNA. The small subunit hosts the decoding center, where codon–anticodon interactions are checked to ensure fidelity during translation.

Functionally, the small subunit binds mRNA and positions the start codon for initiation. In bacteria, initiation often depends on base pairing between the mRNA’s Shine-Dalgarno sequence and the 16S rRNA to align the start codon with the initiator tRNA. In eukaryotes, initiation is typically cap-dependent, with the small subunit scanning the mRNA to locate the start codon in a context defined by surrounding sequence elements (such as the Kozak consensus).

Beyond translation, variants of the term may refer to specific subsystems or processing complexes involved in ribosome biogenesis, such as the small subunit processome in eukaryotes, which participates in early rRNA processing steps before subunit assembly.
