Proximaltodistal, commonly written as proximodistal, refers to the axis or patterning that runs from the proximal (closest to the body) to the distal (farthest from the body) part of a structure, especially in limbs. The term derives from Latin proximus and distalis, used to describe developmental gradients along the limb bud and other appendages.

In embryology, proximodistal patterning governs segmentation and differentiation along the limb axis. Limb development progresses along a proximodistal gradient regulated by signaling centers such as the apical ectodermal ridge (AER) and the zone of polarizing activity (ZPA). Distal identities are promoted by FGF signaling from the AER, maintaining growth and distal fate; proximal identities are promoted by retinoic acid signaling and Meis1/Meis2 expression, establishing a proximal domain. The stylopod (proximal limb segment, such as the humerus or femur) forms closest to the body, followed by the zeugopod (radius/ulna), and finally the autopod (carpals/metacarpals and digits). Disruptions to proximodistal signaling can result in limb malformations, such as loss of distal elements or aberrant proximalization, and are a focus of developmental biology and teratology.

Beyond limbs, proximodistal concepts appear in organ and tissue patterning where structures extend or differentiate along a proximal-to-distal axis, though the limb axis is the most well characterized example.
