Protoeukaryotes are a hypothetical group of primitive organisms considered to be ancestral to all eukaryotes. The concept of protoeukaryotes is central to understanding the origin of eukaryotic cells, a major event in the history of life. These organisms are thought to have possessed some, but not all, of the characteristic features of modern eukaryotes. Key aspects of their evolutionary trajectory involve the development of internal membrane-bound organelles and a more complex cytoskeleton.

The prevailing theory regarding protoeukaryote evolution is endosymbiosis. It is widely believed that the mitochondria and, in some lineages, chloroplasts, originated from free-living prokaryotes (likely alphaproteobacteria and cyanobacteria, respectively) that were engulfed by a host cell. This host cell is considered a protoeukaryote. The engulfment led to a symbiotic relationship where the prokaryotic endosymbionts provided energy, eventually evolving into the organelles we see today.

Another significant evolutionary step for protoeukaryotes was the development of a nucleus. While not enclosed by a double membrane in the earliest stages, the genetic material likely began to be sequestered and organized in a more complex manner than in prokaryotes. The evolution of a more dynamic cytoskeleton also played a crucial role, enabling phagocytosis (the engulfment of other cells) and intracellular transport, which facilitated the endosymbiotic events. The exact nature and lineage of the protoeukaryote host remain subjects of ongoing research and debate.
