Prion protein fragments are pieces of the prion protein (PrP). The normal cellular form of the prion protein, PrPC, is a glycoprotein found on the surface of cells, particularly neurons. Prions are infectious agents composed solely of protein, which can cause fatal neurodegenerative diseases known as transmissible spongiform encephalopathies (TSEs). These diseases include Creutzfeldt-Jakob disease (CJD) in humans, scrapie in sheep, and bovine spongiform encephalopathy (BSE) in cattle.

The key to prion infectivity lies in the misfolding of PrPC into an abnormal, infectious isoform called PrPSc. PrPSc has a different three-dimensional structure and is resistant to degradation. When PrPSc encounters PrPC, it can induce the normal protein to misfold into the abnormal form, leading to a chain reaction.

Prion protein fragments can arise during the normal processing of PrPC or through pathological events associated with prion diseases. Proteolytic cleavage of PrPC can generate various fragments. Some fragments, particularly those that are misfolded or aggregate, may play a role in prion propagation and pathogenesis. Research into these fragments is ongoing, aiming to understand their contribution to disease mechanisms and to develop diagnostic tools or therapeutic strategies. The study of these fragments is crucial for understanding the complex biology of prions and the diseases they cause.
