Pitkälukusekvensointi, also known as long-read sequencing, is a DNA sequencing technology that can generate much longer contiguous sequences of DNA, or reads, compared to traditional short-read sequencing methods. While short-read sequencing typically produces reads of a few hundred base pairs, long-read sequencing can generate reads ranging from thousands to millions of base pairs.

This capability is particularly advantageous for assembling complex genomes. In genomes with repetitive regions or structural variations, short reads can be ambiguous, making it difficult to accurately piece together the complete DNA sequence. Long reads, by spanning these challenging regions, provide more context and reduce ambiguity, leading to more contiguous and accurate genome assemblies. This improved assembly is crucial for identifying genetic variations, understanding genome structure, and studying complex biological processes.

Several technologies enable long-read sequencing, with PacBio (Pacific Biosciences) and Oxford Nanopore Technologies being prominent examples. PacBio's SMRT (Single Molecule, Real-Time) sequencing technology involves observing DNA polymerase activity in real time as it synthesizes a new DNA strand. Oxford Nanopore's technology, on the other hand, detects changes in electrical current as a DNA strand passes through a nanopore. Both methods have contributed significantly to advancements in genomics research, enabling the sequencing of previously difficult-to-assemble genomes and facilitating discoveries in areas such as transcriptomics, epigenomics, and metagenomics. The higher cost and, in some cases, higher error rates compared to short-read sequencing are considerations, but ongoing technological improvements are steadily addressing these challenges.
