One common approach involves serial dilution, where a sample containing cells is progressively diluted, and then plated onto a suitable growth medium. This increases the probability that individual cells are separated from each other. When these diluted samples are incubated, each cell that lands on an agar plate or in liquid culture can divide and form a colony or a culture, respectively. If the initial isolation was done carefully, each colony or culture originates from a single cell and therefore represents a clone.

Another technique is the use of microcapillaries or micromanipulators. This allows for the physical isolation of a single cell under microscopic observation. The isolated cell is then transferred to a nutrient-rich medium where it can divide and proliferate. This method offers a high degree of precision in selecting the starting cell.

These methods are crucial for various applications, including the study of gene function, the production of monoclonal antibodies, and the genetic engineering of microorganisms. By ensuring that a population is derived from a single ancestral cell, researchers can eliminate variability that might arise from mixed populations, leading to more reliable experimental results.
