Lipofiilsed, or lipophilised, is an adjective used in chemistry and pharmacology to describe a molecule that has been modified to increase its lipophilicity, or affinity for fats and nonpolar environments. The term derives from lipophilic, with the British English suffix -ised indicating a past modification.

Etymology and meaning: The root lipos (Greek for fat) and philos (loving) form lipophilic, while lipifi ls ed reflects a process of making a compound more fat-loving. In practice, lipophilization is the deliberate introduction of nonpolar or lipid-compatible groups to a molecule to shift its solubility and distribution properties.

Mechanisms and methods: Lipophilization can be achieved through several chemical strategies, such as adding alkyl or aryl chains, esterification of hydroxyl groups, amide or carbamate formation with fatty acyl moieties, or conjugation with lipid carriers like fatty acids, triglycerides, or phospholipids. The result is an increased partition coefficient (log P), typically accompanied by reduced aqueous solubility. Lipophilization can also be used in the design of prodrugs, where a lipophilic promoiety enhances membrane permeability and is later cleaved in vivo to release an active, more hydrophilic drug.

Applications: In drug design, lipophilized prodrugs and lipid-based formulations aim to improve oral absorption and tissue distribution. Lipophilization is also used in agrochemicals to elevate uptake in plants or soils, and in material science to modify interactions with biological membranes or lipid phases.

Limitations: Higher lipophilicity can lead to poor solubility in water, non-specific tissue distribution, potential toxicity, and altered metabolism. Balancing lipophilicity with solubility and safety is a key challenge in lipophilization strategies.
